49ers vs Rams TNF Preview: Odds, Prediction, Best Bet

Thursday Night Football is kicking off Week 1 in a place it has never been before. The San Francisco 49ers and Los Angeles Rams open the 2026 season on Thursday, September 10 at 8:35 p.m. ET at the Melbourne Cricket Ground in Australia, a Netflix-exclusive broadcast marking the NFL’s first-ever regular-season game Down Under. The Rams are the designated home team, but with roughly 10,000 miles between this stadium and either team’s actual building, this is about as neutral as neutral sites get. Both clubs made deep playoff runs last season before falling to Seattle, and both arrive banged up in different ways, setting up a genuinely intriguing opener despite the unusual setting.

How the Odds Are Shaping Up for the Melbourne Opener

Los Angeles opened as roughly 2.5-to-3-point favorites and the number has crept up to Rams -3.5 at most books, with the total sitting at 48 (down slightly from an opening 48.5). The Rams’ moneyline has moved from around -185 to as steep as -200 in some spots, while the 49ers sit in the +154 to +183 range depending on the book.

The line movement toward Los Angeles isn’t hard to explain. San Francisco has piled up significant defensive attrition in the days leading up to kickoff, while the Rams’ offense — even without their best interior pass rusher on defense — is viewed as the more dependable unit heading into an unusual travel spot for both teams. Bettors tracking the number as it shifts throughout the week can check the latest live NFL odds for updated lines before kickoff.

Team Form, Injuries, and the Players Who Actually Suit Up

The single biggest storyline of the week is who won’t be on the field: Rams defensive tackle Aaron Donald, who came out of retirement last month, will not travel to Australia. Coach Sean McVay made the call himself, citing the grueling flight and his preference not to rush Donald’s return. It’s a coaching decision, not an injury designation, and it removes arguably the most disruptive interior force in football from LA’s front for one week. The rest of the Rams’ key pieces are trending in the right direction — defensive end Myles Garrett is dealing with a knee issue but is expected to play, and star receiver Puka Nacua (core) is also expected to suit up despite being banged up.

San Francisco’s injury report reads heavier. Pass rusher Nick Bosa (knee) and star tight end George Kittle (Achilles) are both expected to play, and Christian McCaffrey is a full go, which matters enormously for an offense that leans on him in every phase. But the 49ers lost depth at receiver before the season even started — Ricky Pearsall and Christian Kirk are both out for the year on injured reserve — which is part of why Mike Evans was brought in and will make his 49ers debut in Melbourne. San Francisco also took a real hit up front this week: defensive tackle Alfred Collins suffered a season-ending torn patellar tendon in practice, thinning out a defensive line that will already be without a full offseason together.

Preseason form slightly favors the Rams on paper — Los Angeles went a perfect 3-0 (wins over the Chargers, Saints, and Chiefs), while San Francisco went 2-1, including a blowout win over the Chargers but a loss to Tennessee. Preseason results rarely predict Week 1 outcomes, but the Rams do appear to be entering the year with more continuity on both sides of the ball.

Two Teams Still Chasing Last January

Both franchises know exactly what’s at stake beyond Week 1 bragging rights. The Rams’ season ended with a 31-27 loss at Seattle in the playoffs, while the 49ers were routed 41-6 in Seattle in mid-January. Neither team wants the Seahawks to be the measuring stick again, and a statement win in Australia — even a quirky, jet-lagged, time-zone-scrambled one — would be a nice way to open the year against a familiar division rival.

Prediction and Best Bet

Take away Aaron Donald and San Francisco’s offensive line gets a real break against a Rams front that’s still plenty talented but noticeably less scary up the middle. That should help Christian McCaffrey find room to work and give Mike Evans a smoother introduction to his new team. But the Rams still have the more complete roster, a superior quarterback situation, and Myles Garrett rushing off the edge to make life difficult for San Francisco’s line all night. Expect a competitive, back-and-forth game that comes down to the fourth quarter.

  • Prediction: Los Angeles Rams 27, San Francisco 49ers 23
  • Best Bet: Rams -3.5

The number has moved a full point in Los Angeles’ favor since it opened, and for good reason — the Rams are the healthier, more complete team even with Donald sidelined, and a four-point cushion feels appropriate for a team that should control most of this game without ever fully pulling away.
